目的 探讨裸花紫珠叶抗肿瘤活性部位的提取工艺。方法 采用MTT比色法对裸花紫珠叶醇提物的不同溶剂萃取物进行抗肿瘤活性测定。选择料液比、提取次数、提取时间、乙醇浓度作为影响因素,以总黄酮含量为指标,采用Box-Behnken设计优化活性部位的提取工艺。结果 乙酸乙酯萃取部位(EAE)对人乳腺癌MCF-7细胞、卵巢癌OVCAR-3细胞和宫颈癌C-33A细胞的增殖具有抑制作用,其IC50值分别为(38.96±2.42)μg/mL、(5.15±3.07)μg/mL、(26.33±1.46)μg/mL(72 h)。活性部位EAE最佳提取工艺为:料液比为1∶20,提取次数为2次,提取时间为2 h,乙醇浓度为70%。结论 裸花紫珠叶乙酸乙酯萃取部位具有一定的抗肿瘤活性,其提取工艺经Box-Behnken设计优化后具有更高的提取率,且工艺稳定,可预测性好,值得推广应用。
Abstract:Objective To explore the extractive technology of active fraction from Callicarpa nudiflora Hook. et Arn. leaves on anti-tumor.Methods The MTT assay was used to determine the anti-tumor activities of different solvent extracts from Callicarpa nudiflora Hook. et Arn. leaves.Total flavonoid content was used as an indicator,and solid-liquid ratio,extract frequency,extract time and ethanol concentration were selected as influence factors to optimize the extractive technology of the active fraction by Box-Behnken design.Results The ethyl acetate extract(EAE) had inhibit activity on MCF-7 cell[IC50=(38.96±2.42) μg/mL],OVCAR-3 cell[IC50=(5.15±3.07) μg/mL]and C-33A cell[IC50=(26.33±1.46) μg/mL]in 72 h,respectively.The optimum extractive technology of EAE was confirmed as solid-liquid ratio was 1:20,extract frequency was 2 times,extract time was 2 h and ethanol concentration 70%.Conclusion The ethyl acetate extract of Callicarpa nudiflora Hook. et Arn. leaves have a certain anti-tumor activity,and the extractive technology after optimized by Box-Behnken design have higher extractive rate,more stabilizer,and better predictability that worth to the popularization and application.
